Strain Calibration

Convert Growth Profiler green values from image analysis into OD with a three-term power curve. Fitting runs in your browser. You can save fittings locally in this browser; nothing is stored on the server.

Calibration instructions

  1. Prepare 10 ml cell suspension with OD 32.
  2. Dilute 0.5 ml of this suspension into 9.5 ml medium/buffer/water to make OD 1.6.
  3. Prepare the dilution series in wells C3–C10 and D3–D10.
  4. Optionally prepare a second strain or duplicate in E3–F10.
  5. Run the Growth Profiler at 4 ms, 250 rpm, 2-minute interval, using at least 3 photos.
  6. Analyze with the image analysis software.
  7. Copy the relevant green values into the table below.
  8. Run the fitting.

This tool models the C3–D10 series (16 wells). E3–F10 is lab guidance only.

Calibration data

Adjust OD only if needed. Green value is the main input.
